Test Sample Preparation:

  1. Mix 10 ml of liquid component with 0.52 g of solid component.
  2. Mixed components were left together for 3 hours before use.
  3. Test products for testing were prepared by adding 30 ml of mixed components in 1L of water.

EN 14476:2019-10 – Quantitative Virucidal Suspension Test for Disinfectant

Test Details:

  • Test Organism: Poliovirus (Fridrich Loeffler Institut (RVB-1260))
  • Test Specimen: Disinfectant STREAM (2023:2380)
  • Test Inoculum: 1 part of test virus into 9 parts of test mixture (8 parts test specimen + 1 part of interfering substance)
  • Contact Time: 30 min
  • Test Temperature: 15°C ± 1°C

Outline of Test Method

  • 1 part of test virus (Poliovirus) was inoculated separately into 9 parts of control (hard water + interfering substance) specimen and 9 parts of test (Disinfectant + interfering substance).
  • The above mixture was pulse vortexed and incubated at a specified temperature (15°C ± 1°C) for 30 min.
  • After the specified contact time (30 min), 0.5 ml of the mixture was added to 4.5 ml of ice-cold medium for neutralization of disinfectant.
  • The neutralized suspension was subjected to serial dilution (1:10) using maintenance medium.
  • Virus recovery from the control and test sample was done by adding the serially diluted sample to cells in a 96-well plate for TCID50 determination.
